The anime's setting is a fictional Japanese city named Sugomori City. One day, Noriko Sonosaki tells her classmate Katsuhira Agata, "You have been selected to be a Kiznaiver." The Kizuna System, which allows Katsuhira to share his wounds, connects him to the classmates whose lives and personalities completely differ from his. The Kizuna System is an incomplete system for the implementation of world peace that connects people through wounds. All those who are connected to this system are called Kiznaivers. When one Kiznaiver is wounded, the system divides and transmits the wound among the other Kiznaivers. Sugomori City is built on reclaimed land, but as the years go by, the city's population is decreasing. The story is set in this town where Katsuhira and the others live.

E:60 covers stories that relate to both American and international sports. Reporters from the network interview those surrounding the stories, and they also discuss what was involved in covering the stories. Many of the stories' subjects are of a serious nature, such as a story featured on the premiere show about Jason Ray, the student who portrayed the North Carolina Tar Heels' mascot Ramses, being killed after he was struck by a car. In each game two members of the audience are selected at random, one from each team, indicated by a light in front of them which remains illuminated when all the other team members' lights have gone off. The games involve contestants competing in acts of bravery, style and talent, some of which are designed to humiliate the contestants, especially an assault course which was introduced with the Italian version in 2010, and the Finale which is a water tank game.

Back Of The Y previously known as Back Of The Y Masterpiece Television is a cult New Zealand TV series, created by Chris Stapp and Matt Heath. Featuring such memorable characters as Randy Campbell, Danny Parker, Dick Johansonson, Cindy Cockburn and Spanners Watson, the show glorified stunts and the consumption of copious amounts of alcohol. The Hollywood special episode featured Randy Campbell jumping off the back of the Y in the famous Hollywood sign. Back of the Y is centred around a mock live TV show, featuring segments such as Randy Campbell's Extreme Stunts and "Cunstables". The show was low budget and apparently filmed with amateur equipment and edited on home computers. The show description at the start of the first episode references one of the cast members and sets the agenda for the rest of the show "Phil Bruff had a dream. To make a New Zealand TV show that wasn't complete shit. He failed... Miserably!!!" The show consisted of 7 episodes of Back Of The Y Masterpiece Television and The Back Of The Y Goes To Hollywood Special. In addition to being screened upon New Zealand television, the show has been given airtime on MTV2 in the UK and on Channel V in Australia. They are also responsible for 'That Cunt Rabbit', Heck and Balls Of Steel.
